Pyloric ligations induce accumulation of acid and pepsin, which leads to autodigestion of gastric mucosa and ulceration. Some studies administer pentagastrin or histamine dihydrochloride subcutaneously.

After the stomach content is collected, the volume of gastric juice is recorded and the concentration of free acid can be calculated and analyzed.
